Decarelin®
Generic name: Triptorelin acetate
Dosage form: Vial containing 100 micrograms of triptorelin acetate in 1 mL of solution for injection
Pharmacological class: Gonadotropin-releasing hormone (GnRH) agonist
Indications: This medication is used as an adjunct treatment for infertility in women undergoing controlled ovarian stimulation protocols.
Decarelin® is prescribed in infertility treatment as part of Assisted Reproductive Technology (ART) protocols.
It belongs to the pharmacologicalclass of gonadotropin-releasing hormone (GnRH) agonists and prevents the premature surge of luteinizing hormone (LH), thereby inhibiting early ovulation during the menstrual cycle. Once the follicles are ready for retrieval, ovulation-inducing agents are administered to trigger oocyte release.
Decarelin® is used in controlled ovarian stimulation protocols for infertility treatment by preventing premature luteinizing hormone (LH) surge.
Vial containing 1 mL of solution for injection (0.1 mg/mL)
Each vial contains 100 micrograms of triptorelin acetate, equivalent to 95.6 micrograms of triptorelin.
Decarelin® is supplied as a ready-to-use solution for injection and does not require any preparation.
Decarelin® should be prescribed by a specialist physician.
Treatment is initiated either in the early follicular phase (day 2 or 3 of the menstrual cycle) or in the mid-luteal phase (days 21 to 23 of the cycle, corresponding to 5–7 days before the onset of menstruation), and continues until the day of ovulation triggering.
Decarelin® is administered as a daily subcutaneous injection, approximately less than 5 cm away from the navel. To minimize local hypersensitivity reactions, the injection site on the abdomen should be rotated daily. The selected site should not be painful, bruised, erythematous, or affected by any skin lesion.
- Clean the injection site using an alcohol swab and allow it to dry.
- Gently pinch the skin with one hand.
- Insert the needle at a 90-degree angle (45 degrees in lean individuals), inject the full content of the syringe, and release the skin after counting to three.
- Remove the needle while applying gentle pressure with an alcohol pad.
Note: The vial included in the Decarelin® package, as well as the syringe and needle obtained from the pharmacy, are intended for single use only. Dispose them appropriately after use.
If a dose of Decarelin®is missed, consult your physician.
In case of administration of a higher-than-recommended dose, inform your physician.
- Negative pregnancy test prior to initiating triptorelin therapy
- Serum estradiol levels and ultrasound monitoring (to assess follicular growth, size, and number)
- Signs and symptoms of ovarian hyperstimulation syndrome (OHSS)
- Signs of allergic reactions within 30 minutes post-injection
- Blood glucose or HbA1c, electrolytes, serum lipids, blood pressure, and ECG
- Emergence of cardiovascular symptoms
- Emergence of psychiatric symptoms
- History of hypersensitivity reactions to triptorelin or any of the formulation components
- History of hypersensitivity reactions to other gonadotropin-releasing hormone (GnRH) agonists or other hormones with similar activity
- Pregnancy or lactation
- Unexplained vaginal bleeding
During and after treatment with triptorelin, ovarian hyperstimulation syndrome (OHSS) may occur. Although this is a known potential complication of ovarian stimulation in assisted reproductive technology (ART) protocols, patients should be closely monitored for symptoms such as moderate to severe abdominal pain, abdominal distension and firmness, nausea, and vomiting. In case of such symptoms, immediate medical intervention under physician supervision is required.
Patients with reduced bone mineral density or urinary tract obstruction should inform their physician before initiating treatment.
The use of triptorelin during pregnancy and lactation is contraindicated.
Pregnancy must be ruled out prior to initiating treatment.
As with all medications, Decarelin® may cause adverse effects, although not all patients will experience them. Possible adverse reactions include:
- Hypersensitivity reactions
- Ovarian hyperstimulation syndrome (OHSS), with symptoms including moderate to severe abdominal pain, abdominal distension and firmness, nausea, and vomiting
Patients should consult their physician or pharmacist regarding the concomitant use of triptorelin with other medications or supplements.
Each package contains 5 vials of Decarelin®.
- Keep out of the sight and reach of children.
- Store at 2–8°C, protected from light and moisture.
- Do not freeze.
- The product may be used until the end of the month indicated in the expiration date on the packaging. Do not use or store expired medication.
- Keep the medication in its original packaging until use.
